Abstract

The objective of this study was to assess the effect of Xenical on plasma concentrations of xenobiotics in patients with rheumatoid arthritis (RA) and rheumatoid arthritis (RA/TRAFlu) by a single dose and a placebo. The pharmacokinetics of xenobiotics was assessed in two groups of patients with rheumatoid arthritis (RA) and RA/TRAFlu. In the second group, plasma concentrations of xenobiotics were measured in patients with RA. A total of 50 patients were enrolled in the pharmacokinetic study. All the patients were given either a single oral dose of xenobiotics or a placebo. At 48 h after the drug administration, a single dose of Xenical (3.5 mg) had an area under the plasma concentration-time curve (AUC) of the drug similar to that of the placebo, but no effect was observed. At 48 h after the drug administration, plasma concentrations of xenobiotics were within the therapeutic range for all patients. Pharmacokinetic analysis of plasma concentrations of xenobiotics revealed that Xenical had no significant effect on the pharmacokinetics of the drugs in patients with RA. The pharmacokinetic data of xenobiotics administered to healthy volunteers did not change significantly in comparison to the plasma concentrations of the drugs in patients with RA and RA/TRAFlu. The pharmacokinetic data of xenobiotics administered to healthy volunteers did not change in comparison to the plasma concentrations of the drugs in patients with RA and RA/TRAFlu.

Arthritis and other rheumatic diseases are a serious and growing cause of disability and death. The World Health Organization (WHO) estimates that approximately 10 million people in the world suffer from rheumatic diseases, with a lifetime prevalence of more than one million. This figure is in line with the need for effective treatment of rheumatic diseases and their associated diseases. However, despite the increasing recognition of these diseases, they still persist to a considerable extent, leading to lack of effective and safe treatment options.

Rheumatoid arthritis (RA) is a chronic inflammatory disease of the rheumatism that is prevalent in patients and has a high prevalence in the general population. RA is the most common systemic disease in patients with rheumatoid arthritis, with a prevalence of over two million individuals. The main therapeutic targets for the treatment of RA are: anti-inflammatory factors, such as the glucocorticoid and glucoside hormones, and anti-infective factors. The main indications for RA treatment are systemic corticosteroids and glucocorticoids.
